    Semax and Selank are often grouped together as “Russian nootropic peptides,” and they do share a research lineage, but their proposed mechanisms and researched use cases differ. This post compares what’s published about each.

    Shared Origins, Different Targets

    Both peptides were developed at the Institute of Molecular Genetics of the Russian Academy of Sciences and are registered pharmaceutical drugs in Russia, but neither is FDA-approved in the United States. Semax is a synthetic heptapeptide derived from ACTH(4-10), a fragment of adrenocorticotropic hormone, modified with a Pro-Gly-Pro tail that extends its stability without retaining ACTH’s adrenal-stimulating activity. Semax has been registered in Russia since 1994 for indications including ischemic stroke recovery, cognitive impairment, optic nerve atrophy, and attention difficulties, and was added to Russia’s List of Vital and Essential Drugs in 2011.

    Selank is a synthetic analog of tuftsin, an immunomodulatory peptide, engineered with an added Pro-Gly-Pro sequence for similar stability reasons. Its research profile centers on anxiety and mood regulation rather than stroke recovery.

    What Published Research Explores

    Semax research has most consistently reported upregulation of BDNF (brain-derived neurotrophic factor) and NGF (nerve growth factor) in hippocampal and cortical tissue in animal studies, along with changes in dopamine and serotonin turnover. A genome-wide transcriptional analysis of Semax in a rat brain ischemia model found effects on gene expression related to immune and vascular systems in brain tissue, offering a broader look at its mechanism beyond single-pathway studies.

    Selank’s primary proposed CNS mechanism is positive modulation of GABA-A receptor sensitivity — increasing the receptor’s responsiveness to the body’s own GABA rather than acting as a direct agonist — alongside reported rapid elevation of hippocampal BDNF in animal studies, which researchers have linked to potential effects on neuroplasticity.

    What’s Still Unknown

    Most of the mechanistic detail for both peptides comes from Russian-language and animal-model research; independent replication in Western, peer-reviewed literature is comparatively limited for both. Semax is also currently under FDA review: the Pharmacy Compounding Advisory Committee’s July 2026 meeting materials flag anticoagulant and amphetamine-potentiation signals associated with Semax as an open safety question, alongside the agency’s proposal not to add it to the 503A bulks list.

    Common Misunderstandings

    Being an approved drug in Russia is not equivalent to FDA approval or to a demonstrated safety and efficacy profile under US regulatory standards — different regulatory bodies apply different evidentiary thresholds. Grouping Semax and Selank together as interchangeable “nootropics” also glosses over that one targets neurotrophic/monoaminergic pathways studied mainly for stroke and cognition, while the other targets GABA-A modulation studied mainly for anxiety.
